MGF505-3R inhibited IFN-β activity through multiple sites. A-E HEK293T cells were cultured overnight in 24-well plates, and increasing doses of HA-MGF505-3R plasmid (50 ng, 100 ng, 200 ng) were cotransfected with the IFN-β-Luc plasmid (100 ng), pRL-TK plasmid (50 ng) and signaling molecule plasmids (Flag-cGAS, Flag-STING, Flag-TBK1, Flag-IKKε, Flag-IRF3) (200 ng) for 24 h. “+” represents transfected, “-” represents nontransfected. For all cell lysates, the target proteins were detected by dual-luciferase reporter assays and western blotting. Quantitative analysis of protein blots was performed using ImageJ software. All data from 3 independent experiments are expressed as the mean ± SD. *P<0.05, **P<0.01, ***P<0.001
