MGF505-3R knockdown enhanced IFN-β and IL-1β production. PAMs were transfected with siMGF505-3R-259 or nontargeting control siRNA for 24 h followed by infection with ASFV (MOI=1) for 12 h or 24 h. Cells were collected at the indicated times. A-D Endogenous MGF505-3R, ISG15, ISG56, and IFNB1 mRNA levels were assessed by qRT–PCR assay. E ELISA was used to assess secretion of the cytokine IL-1β in the cell supernatant. F-H Western blotting was used to assess cellular or viral protein expression levels and quantitative analysis of protein blots using ImageJ software. “+” represents transfected, “-” represents nontransfected. All experiments were repeated independently at least thrice. All data from 3 independent experiments are expressed as the mean ± SD. *{P<0.05}, **{P<0.01}, ***{P<0.001}
